Dosing & Reconstitution Guide
Educational guide for reconstitution and daily dosing
Standard / Gradual Approach (3 mL = ~3.333 mg/mL)
| Week | Daily Dose (mcg) | Units (per injection) (mL) |
|---|---|---|
| Weeks 1–4 | 300 mcg | 9 units (0.09 mL) |
| Weeks 5–12 | 500 mcg | 15 units (0.15 mL) |
Frequency: Inject once daily subcutaneously (typically in the morning on an empty stomach). This schedule uses the largest practical dilution (3.0 mL). For the 9-unit measurement, consider a 30- or 50-unit insulin syringe for improved readability. Rotate injection sites (abdomen, thighs, upper arms) to minimize local irritation.
Reconstitution Steps
- Draw 3.0 mL bacteriostatic water with a sterile syringe.
- Inject slowly down the vial wall; avoid foaming.
- Gently swirl/roll until dissolved (do not shake).
- Label and refrigerate at 2–8 °C (35.6–46.4 °F), protected from light.
Storage note: These temperatures are general laboratory handling guidance, not a validated use-by period for this AOD-9604 preparation. Follow the exact product instructions if they differ.[17][19]
Supplies Needed
Plan based on an 8–12 week daily protocol with gradual titration (300 mcg Weeks 1–4, 500 mcg Weeks 5+).

-
Alcohol Swabs: One for the vial stopper + one for the injection site each day.
- Per week: 14 swabs (2/day)
- 8 weeks: 112 swabs - 2 boxes required (100 swabs each)
- 12 weeks: 168 swabs - 2 boxes required (100 swabs each)
AOD-9604 Vial and Research Context
- Reconstitute: Add 3.0 mL bacteriostatic water → ~3.333 mg/mL (3333 mcg/mL) concentration.
- Daily range: 300–500 mcg once daily (gradual titration).
- Easy measuring: At 3.333 mg/mL, 1 unit = 0.01 mL ≈ 33.33 mcg on a U‑100 insulin syringe.
- Storage (general laboratory handling): Lyophilized: freeze at −20 °C (−4 °F); after reconstitution, refrigerate at 2–8 °C (35.6–46.4 °F); avoid freeze–thaw cycles. See the formulation limits in Storage Instructions below.[17]
AOD-9604 is a modified fragment based on the C-terminal region of human growth hormone. Animal research investigated its effects on lipid metabolism.[1][4] Human safety findings summarized oral and intravenous studies; they do not establish a placebo-like safety profile for subcutaneous administration of this 10 mg vial preparation.[2] FDA’s 2024 review found no human subcutaneous clinical data and insufficient evidence of effectiveness for obesity.[3]
Protocol Overview
Concise summary of the once‑daily regimen.
- Goal: Support reduction of fat mass and enhance fat oxidation over time[1][4].
- Schedule: Daily subcutaneous injections for 8–12 weeks.
- Dose Range: 300–500 mcg daily with gradual titration.
- Reconstitution: 3.0 mL per 10 mg vial (~3.333 mg/mL) for accurate unit measurements.
- Storage (general laboratory handling): Lyophilized frozen; reconstituted refrigerated; avoid repeated freeze–thaw. Product-specific directions take precedence.[17]
Dosing Protocol
Suggested daily titration approach.
- Start: 300 mcg daily for Weeks 1–4; increase to 500 mcg for Weeks 5–12 as tolerated.
- Target: 500 mcg daily by Week 5 and maintain through cycle completion.
- Frequency: Once per day (subcutaneous).
- Cycle Length: 8–12 weeks.
- Timing: Morning administration (fasted) is common; rotate injection sites.
Storage Instructions
General peptide guides provide a laboratory handling reference, not a validated shelf life for this AOD-9604 10 mg preparation. The exact product’s documented instructions take precedence.
- Dry powder: Keep lyophilized peptide tightly capped and dry. General laboratory guidance uses a freezer at −4 °F (−20 °C) for frozen storage and advises avoiding repeated freeze–thaw cycles.[17]
- After mixing: Sigma’s general peptide guide describes short-term refrigeration around 39 °F (4 °C), within the refrigerated range shown above. This is not a guarantee of stability for every peptide solution. Keep the vial capped and avoid repeated freeze–thaw cycles.[17]
- Light and handling: Protect laboratory peptide preparations from direct light and minimize unnecessary handling.[18]
No validated use-by period is assigned here. Stability testing needs to match the actual formulation and reconstituted concentration. Refrigeration and bacteriostatic water do not establish this vial’s potency, sterility or a 28-day usable life.[19]
AOD-9604 10 mg Dosage: Calculation and Evidence
AOD-9604 10 mg dosage arithmetic depends on both vial mass and final volume. With 10 mg in 3 mL, the calculated concentration is about 3.333 mg/mL. At that concentration, each U-100 unit contains about 33.33 mcg.
AOD 9604 dosage calculations cannot establish a treatment protocol. The human safety literature summarized oral and intravenous studies. Those findings do not validate subcutaneous administration of a commercially supplied vial.[2][3]
A Growth Hormone Fragment, Not Full-Length Growth Hormone
AOD-9604 is a modified fragment based on the C-terminal region of human growth hormone. Researchers investigated its metabolic effects, but the fragment’s proposed actions should not be treated as the established actions of full-length growth hormone.[1]
In obese mice and mice lacking the beta-3 adrenergic receptor, investigators examined lipid metabolism and the receptor’s role in the response. That experiment provides animal-model evidence, not proof that an AOD-9604 injection produces weight loss in people.[4]
Human Findings and Subcutaneous Safety Limits
The 2013 human safety report summarized six randomized trials involving oral or intravenous AOD-9604. It reported no significant increase in IGF-I and generally similar adverse-event findings between study groups under those tested conditions. It did not establish a safety profile for this subcutaneous preparation.[2]
FDA’s 2024 compounding review found no human subcutaneous clinical data and insufficient evidence of effectiveness for obesity. A selected result from an earlier trial cannot resolve those broader limitations.[3]
FDA also identifies limited safety information, possible immunogenicity and impurity-related concerns, and serious adverse events that may be associated with AOD-9604. The agency notes that causality is unclear. That is a reason for caution, not proof that every reported event was caused by the compound.[15]
How to Compare AOD-9604 Studies
Check the route before interpreting a result. Oral exposure, intravenous exposure, and a subcutaneous vial protocol are different interventions. The measured endpoint matters too: a safety study is not automatically a demonstration of weight-loss effectiveness.[2][3]
Descriptions such as fat breakdown or metabolic support can summarize a research question. They should not turn a proposed mechanism into a promised result for the person reading a dosage chart.
Reconstitution and U-100 Measurement Checks
U-100 units are volume markings, not peptide international units. One unit is 0.01 mL. A concentration belongs to a particular final volume, so the same syringe marking will contain a different mass after a different dilution.
Check the syringe’s capacity and graduations separately from the device used to measure the full preparation volume. A vial’s nominal capacity does not independently verify solubility, sterile preparation, or compatibility with bacteriostatic water.
CDC’s general injection-safety guidance calls for new sterile equipment when accessing a vial and disposal of used needles and syringes. It does not approve an AOD-9604 formulation or protocol.[16]
