One-Vial Calculation Using the Published 20 mcg Daily Amount
One-Vial Calculation Using the Published 20 mcg Daily Amount
| Research Period | Amount per Administration | U-100 Units (mL) |
|---|---|---|
| 37 daily administrations (one-vial arithmetic) | 20 mcg | 8 units (0.08 mL) |
Frequency: Once daily in the cited human trials and approved labeling.[1][2][3][4]
Research reference only: The 20 mcg amount is supported by human trials. The 37-administration period is only the number of complete 20 mcg calculations in one 750 mcg vial; it is not a trial duration, stability period, or recommendation.
Reconstitution Steps
- Clean the vial stopper and diluent stopper with alcohol; allow both to dry.
- Draw exactly 3.0 mL of bacteriostatic water with a new sterile syringe, then inject it slowly down the inside wall of the vial.
- Gently swirl or roll until dissolved. Do not shake.
- Label the vial with the compound, concentration, and reconstitution date. Refrigerate at 2–8 °C (36–46 °F) and protect from light.
Storage note: No compound-specific post-reconstitution stability or discard period has been established for this worked research-vial preparation. Follow the lot-specific product documentation for any shorter limit.
Supplies Needed
This list covers the 37 complete 20 mcg arithmetic amounts contained in one 750 mcg vial.
- Peptide Vials (Teriparatide, 750 mcg each):
- One-vial calculation: 1 vial
- Insulin Syringes (U-100):
- 37 calculated administrations: 37 syringes
- Bacteriostatic Water (10 mL bottle):
- One vial at 3.0 mL: 3.0 mL total
- Alcohol Swabs:
- 37 calculated administrations at 2 swabs each: 74 swabs
Sharps safety: Place used needles and syringes immediately in an approved sharps container.
Teriparatide Vial and Research Context
- Reconstitute: Add 3.0 mL bacteriostatic water for a worked concentration of 250 mcg/mL.
- Vial contents: 750 mcg Teriparatide.
- U-100 conversion: 1 unit = 0.01 mL = 2.5 mcg.
- Storage: Refrigerate at 2–8 °C and protect from light; approved-pen handling instructions are formulation-specific.
The 750 mcg vial math is not interchangeable with the approved teriparatide pen. The page preserves the published human amount while clearly separating it from research-vial preparation and stability assumptions.[1]
Teriparatide Dosage Evidence
Teriparatide is a parathyroid hormone analog studied extensively in osteoporosis. Randomized trials evaluated 20 mcg subcutaneously once daily in postmenopausal women, men, and glucocorticoid-induced osteoporosis populations.[2][3][4] Other research compared daily, weekly, and cyclic schedules rather than treating them as interchangeable.[5][6]
The approved product is a liquid multidose pen at 250 mcg/mL.[1] A 750 mcg lyophilized research vial has different formulation, sterility, excipient, stability, and delivery questions. The table on this page therefore uses the published 20 mcg amount only as a transparent arithmetic reference.
750 mcg Vial Calculation Notes
- 750 mcg divided by 3.0 mL equals 250 mcg/mL.
- One U-100 unit equals 0.01 mL or 2.5 mcg.
- 20 mcg equals 8 units (0.08 mL).
- One vial contains 37 complete 20 mcg calculated amounts with 10 mcg remaining. This is vial arithmetic, not a use-by period.
Human Research and Treatment Sequences
Clinical research has examined teriparatide alone, in combination with denosumab, and in treatment sequences.[7][8][9] Biosimilar and active-comparator trials also used the 20 mcg daily schedule.[10][11] A transdermal study used injected 20 mcg teriparatide as a comparator, illustrating that delivery systems must be evaluated separately.[12]
Safety and Formulation Limits
The official label contains formulation-specific warnings, adverse reactions, handling, and pen instructions.[1] Major osteoporosis guidelines discuss patient selection and monitoring for approved therapy.[13][14] Comparative osteoporosis research should not be used to infer that a third-party vial is clinically equivalent.[15]
